Entwicklung und Validierung der Steigbildmethode zur Differenzierung von ausgewählten Lebensmitteln aus verschiedenen Anbausystemen und Verarbeitungsprozessen

Seit den 1980iger Jahren wächst die Bedeutung der sog. Bildschaffenden Methoden für die Bestimmung der Qualität ökologischer Produkte. Zu diesen Methoden gehört die Biokristallisation, Steigbild und Rundfilter-Chromatographie. Die Ergebnisse dieser Methoden sind Bilder, die anhand definierter Kriterien ausgewertet werden. Bei der Biokristallisation sind es mehr oder weniger geordnete Kristallisationen auf einer Glasplatte, bei dem Steigbild zweidimensionale Strukturen auf Chromatographiepapier. In der Vergangenheit wurden die Bilder von Spezialisten ausgewertet, die nach einer längeren Schulung produktspezifische Kriterien entwickelt hatten.
Im Gegensatz zur Dünnschicht-Chromatographie, wo der einzelne Stoff von der Matrix separiert wird, ist das Ziel beim Steigbild, Strukturen der möglichst ganzen Probe zu erzeugen. Die Methode wurde von Kolisko in den 1929iger Jahren entwickelt, wobei eine Kombination aus Chromatographieprozess und Metallkomplexreaktionen genutzt wurde. Die Firma WALA entwickelte die Methode für die Kontrolle ihrer Produkte und setze Silbernitrat und Eisensulfat ein.
Bisher wurde die Methode qualitativ beschreibend ausgewertet, wobei einzelne Bildelemente und deren Interaktion beschrieben wurden. Deshalb musste für die vorliegende Arbeit Auswertungsmethoden entwickelt werden, mit denen auch eine statistische Bearbeitung der Ergebnisse möglich ist (nominale Unterscheidung von proben anhand der Bilder). Die Methode wurde bisher in einer Reihe von Studien eingesetzt (u.a. die Unterscheidung von Produktionsweisen). Obwohl die Bilder nur qualitativ ausgewertet wurden, konnten geschulte Prüfpersonen Proben aus verschiedenen Anbausystemen anhand der Bilder trennen. Die Ergebnisse wurden aber nicht so dokumentiert, dass sie den Erfordernissen internationaler Standardnormen für Laboratorien genügten. Deshalb mussten für diese Arbeit zunächst die Prozeduren dokumentiert und eine systematische Untersuchung zu den Einflussgrößen durchgeführt werden. Dazu wurde die visuelle Bildauswertung entwickelt und standardisiert. Die visuelle Bildauswertung basiert auf morphologischen Kriterien der Bilder von den untersuchten Weizen- und Möhrenproben. Ein Panel aus geschulten Personen entwickelte dann die Kriterien und legte sie anhand von Referenzbildern fest. Die Bilder der vorliegenden Arbeit wurden mit der einfach beschreibenden Prüfung ausgewertet, wie sie aus der sensorischen Prüfung von Lebensmitteln übernommen werden konnte. Mit geschulten und ungeschulten Prüfpersonen wurden Weizenproben und verschiedene Möhrensäfte mit der sog. Dreiecksprüfung ausgewertet (von ISO 4120).
Alle Laborprozeduren wurden dokumentiert. Mit der Anwendung dieser Prozeduren wurden Vergleichsversuche mit Laboren in Dänemark und Holland (BRAD, LBI) durchgeführt. Die Ergebnisse waren sowohl für Weizen- als auch für Möhrenproben vergleichbar, wobei alle drei Labore zwischen jeweils zwei Proben unterscheiden konnten. Die systematische Untersuchung zu den Einflussgrößen zeigte, dass das Unterscheidungsvermögen der Methode vor allem von den klimatischen Bedingungen während der Steigphasen beeinflusst wird. Auch die Präkonditionierung der Papiere hat einen großen Einfluss, während die Wasserqualität (ultra-filtriert, de-ionisiert, destilliert) eine untergeordnete Bedeutung hat. Für Weizen- und Möhrenproben wurde sowohl die Wiederholbarkeit als auch die Reproduzierbarkeit getestet. Die Unterschiede in den Bildern der verschiedenen Proben waren dabei immer größer als die Variation durch Proben- und Bildwiederholung und das Labor.
Die so charakterisierte Methode wurde auf kodierte Proben von definierten Feldversuchen und auf Marktproben (Paarvergleich von Anbausystemen ökologisch und konventionell) angewandt, wobei als Ergebnis mehr als 90% der Proben mit der einfach beschreibenden Prüfung anhand der Bilder unterschieden werden konnten. Die Auswertung mit der Dreiecksprüfung zeigte, dass sowohl Sorten und Verarbeitungsschritte (Saft) als auch Anbauweisen signifikant getrennt wurden. Darüber hinaus wurde die Methode auch erfolgreich auf Apfelproben angewandt. Weitere Untersuchungen müssen zeigen, ob sich das Potential der Methode, verschiedene Fragen wie die Authentizitätsprüfung von Lebensmitteln verifizieren lassen.

Summary (english)
Since the 1980s the picture developing methods of food quality determination in organic food have become more important. Picture developing methods include such methods as biocrystallisation, capillary dynamolysis and round-filter chromatographic methods. The results of these methods are patterns, which are evaluated using defined criteria. Biocystallisation shows more or less organised crystal patterns on a round glass plate whereas the patterns from the capillary dynamolysis method are 2-dimensional structures on paper. In the past the patterns resulting from these methods have needed to be evaluated by well trained personnel using product specific criteria.
In contrast to thin-layer chromatography, where the compound is separated from the matrix and other substances, the aim of the capillary dynamolysis is to produce structured patterns of the whole juice or extract. The capillary dynamolysis method was originally discovered by Kolisko in the 1920s, where a combination of the capillary analysis and metal-salt-solutions were used. The WALA company applied the method for quality control of their pharmaceuticals. For this they used AgNO3 and introduced FeSO4 to increase the number of form elements as well as to fix the paper. Before the work reported in this paper was undertaken pictures resulting from the above methods had been evaluated qualitatively, describing the different form elements and their interactions. Therefore an evaluation tool had to be developed which allowed a statistical test of the results (nominal differentiation). The capillary dynamolysis method was applied to several different studies for comparison (e.g. farming systems). Although the pictures were only evaluated qualitatively, trained personnel could successfully discriminate samples from different farming systems. However, these results were never documented in a way which met the international standard norms for laboratory methods. Therefore the procedures for these method had to be documented and a systemic investigation of the different influencing factors had to be undertaken. Moreover a visual evaluation method had to be developed and standardised. These tasks are the topic of this Thesis.
The visual evaluation of the patterns concerns different morphological criteria for the wheat and carrot samples. A panel of trained assessors was formed and in a consensus meeting all criteria were discussed, agreed and defined. The pictures are evaluated by simple descriptive tests, which were already developed and applied in sensory testing. Moreover the patterns of the wheat samples (varieties) and the carrot juices were evaluated by untrained assessors in a triangular test which was adopted from sensory analysis (ISO 4120).
All laboratory procedures were documented according to international standard norms. Using these procedures comparative studies were undertaken between three partner laboratories (Biodynamic Research Association, University of Kassel, Louis Bolk Institute) that resulted in comparable patterns for wheat (variability higher) and carrot samples. Moreover the discrimination ability of the 2 different product types was the same in all laboratories. A systemic investigation of the different factors influencing the patterns was carried out. The discrimination ability between the different wheat and carrot samples is mainly influenced by the climate conditions during the rising phases. Also the preconditioning of the paper material alters the differentiation between the samples. However water from different cleaning systems (ultra-filtration, de-ionization, distillation) does not seem to have an influence. For both samples types (wheat, carrots) the method parameters, repeatability, intermediate precision and reproducibility was tested. The differences between the patterns of the pictures from all samples were greater than the variation from the parallel repetition and the laboratory influence.
When the method was applied on coded samples, from defined field trials and from the market samples (pair-wise comparison of organic and conventional farms) more than 90% could be differentiated when the simple descriptive test was applied. When the triangular test was used for the differentiation of the wheat and carrot samples (varieties), carrot juices and carrot samples from different farming systems highly significant results could be achieved.
The method was also successfully applied to apple samples from different farming systems. Further investigations need to be carried out to verify the ability of the method to answervarious questions such as the authenticity test of organic crop products for classification.

Od momentu pojawienia sie na rynku produktow rolnictwa ekologicznego, naukowcy staraja
sie ustalic roznice jakosciowe pomiedzy produktami ekologicznymi a konwencjonalnymi.
Badania jakosci zywnosci odbywaja sie przy zastosowaniu metod analitycznych. W ostatnich
latach na znaczeniu zyskuja rowniez tzw. metody holistyczne, do ktorych naleza m.in. metody
obrazowania: biokrystalizacja oraz chromatografia obrazowa. Wyniki tych metod to wzory
oceniane przy uzyciu zdefiniowanych kryteriow. W przypadku krystalizacji wzory te to
skrystalizowane na szklanej plytce substancje. Podczas chromatografii obrazowej na papierze
chromatograficznym powstaja wzory dwuwymiarowe. W porownaniu z chromatografia
cienkowarstwowa, ktorej celem jest wyizolowanie poszczegolnych substancji z badanej proby,
celem chromatografii obrazowej jest uzyskanie odwzorowania czyli wzoru
charakterystecznego dla badanej proby. Metoda chromatografii obrazowej odkryta zostala
przez Kolisko na poczatku minionego wieku. Kolisko zmodyfikowala metode chromatografii
bibulowej m.in. poprzez zastosowanie roztworow soli metali. Firma WALA zastosowala te
metode do kontroli jakosci surowcow farmaceutycznych. Jako odczynnikow chemicznych
uzyto AgNO3 i FeSO4. Wprowadzenie FeSO4 umozliwilo uwidocznienie wiekszej liczby
elementow we wzorach i ich utrwalenie. Przed rozpoczeciem badan do niniejszej pracy, wzory
oceniano jakosciowo (opis roznych elementow wzorow i ich interakcji). Dlatego tez jednym z
zadan bylo opracowanie takiej metodyki oceny wzorow, aby umozliwic statystyczna ocena
wynikow. Chromatografia obrazowa znalazla zastosowanie w niektorych studiach
porownawczych, badajacych jakosc produktow spozywczych (np. porownanie systemow
uprawy). Z dotychczas opublikowanych badan wynika, ze pomimo wylacznie jakosciowej
oceny wzorow, osoby wyszkolone byly w stanie prawidlowo rozrozniac pomiedzy probami
pochodzacymi z roznych systemow uprawy. Jednakze wyniki te w zadnym z przypadkow nie
zostaly udokumentowane tak jak wymagaja tego wyznaczone w normach standardy, przyjete
dla przeprowadzania metod laboratoryjnych. Ppodjeto sie wiec udokumentowania procedur
laboratoryjnych, oraz przeprowadzenia systematycznych badan czynnikow, mogacych miec
wplyw na wzory i modyfikujacych zdolnosc rozrozniania prob za pomoca metody
chromatografii. Ponadto opracowana i przeprowadzena zostala metoda oceny wizualnej
wzorow, oparta o rozne kryteria morfologiczne, bedace do zaobserwowania we wzorach
chromatogramow wykonanych z prob pszenicy i marchwi. Kryteria oceny zostaly ustalone
przez zespol wyszkolonych osob, poddane dyskusji i jednomyslnie zdefiniowane. Wszystkie
wzory chromatograficzne zamieszczone w tej pracy zostaly ocenione za pomoca testu
opisowego, opracowanego dla badan sensorycznych i ujetego w normie DIN 10964 .
Dodatkowo wzory prob pszenicy (odmiany) oraz proby sokow z marchwi zostaly ocenione za
pomoca testu triangle, stosowanego w analizie sensorycznej i zaadaptowanego dla potrzeb
metody chromatografii obrazowej (ISO 4120). Wszystkie procedury laboratoryjne zostaly
udokumentowane zgodnie z ustalonymi w normach standardami. Badania porownawcze
pomiedzy trzema wspolpracujacymi ze soba laboratoriami (Biodynamic Research Association
DK, Uniwersytet Kassel D, Louis Bolk Institute NL) wykazaly, ze zastosowanie
udokumentowanych procedur prowadzi w kazdym z laboratoriow do porownywalnych wzorow
zarowno dla prob pszenicy jak i dla sokow z marchwi. Ponadto zdolnosc rozrozniania roznych
prob pszenicy i sokow z marchwi za pomoca metody chromatografii obrazowej byla
jednakowa we wszystkich laboratoriach. Systematyczne badania nad czynnikami, mogacymi
miec wplyw na zmiany we wzorach wykazaly, ze glownym czynnikiem modyfikujacym
zdolnosc rozrozniania prob metoda chromatografii sa warunki klimatyczne, panujace w
laboratorium podczas przeprowadzania doswiadczenia. Podobny wplyw wywiera
kondycjonowanie papieru chromatograficznego przed doswiadczeniem, podczas gdy np.
zastosowanie wody z roznych systemow oczyszczania (ultrafiltracja, dejonizacja, destylacja)
nie ma wplywu na zmiany we wzorach, a w zwiazku z tym rowniez na zdolnosc rozrozniania
prob. Takie parametry jak powtarzalnosc wynikow, precyzja metody i odtwarzalnosc wynikow
zostaly dla metody chromatografii obrazowej przetestowane na probach pszenicy (odmiany)
oraz sokow z marchwi. Zastosowanie metody do rozrozniania zakodowanych prob ze
zdefiniowanych doswiadczen polowych oraz z rynku (porownanie par produktow z
ekologicznych i konwencjonalnych gospodarstw rolnych) umozliwilo poprawne
zaklasyfikowanie >90% prob (met. opisowa). Zastosowanie testu triangle do rozrozniania prob
marchwi i pszenicy (odmiany), sokow z marchwi i prob marchwi z roznych systemow uprawy
pozwolilo na uzyskanie znaczacych statystycznie roznic. Dalsze badania sa niezbedne, aby
ustalic przydatnosc metody chromatografii obrazowej do pracy nad roznorodnymi
zagadnieniami, takimi jak autentycznosc pochodzenia produktow spozywczych i ich
klasyfikacja.

DK, Uniwersytet Kassel D, Louis Bolk Institute NL) wykazaly, ze zastosowanie
udokumentowanych procedur prowadzi w kazdym z laboratoriow do porownywalnych wzorow
zarowno dla prob pszenicy jak i dla sokow z marchwi. Ponadto zdolnosc rozrozniania roznych
prob pszenicy i sokow z marchwi za pomoca metody chromatografii obrazowej byla
jednakowa we wszystkich laboratoriach. Systematyczne badania nad czynnikami, mogacymi
miec wplyw na zmiany we wzorach wykazaly, ze glownym czynnikiem modyfikujacym

Bildschaffenden Methoden für die Bestimmung der Qualität ökologischer Produkte. Zu diesen Methoden gehört die Biokristallisation, Steigbild und Rundfilter-Chromatographie. Die Ergebnisse dieser Methoden sind Bilder, die anhand definierter Kriterien ausgewertet werden. Bei der Biokristallisation sind es mehr oder weniger geordnete Kristallisationen auf einer Glasplatte, bei dem Steigbild zweidimensionale Strukturen auf Chromatographiepapier. In der Vergangenheit wurden die Bilder von Spezialisten ausgewertet, die nach einer längeren Schulung produktspezifische Kriterien entwickelt hatten. Im Gegensatz zur Dünnschicht-Chromatographie, wo der einzelne Stoff von der Matrix separiert wird, ist das Ziel beim Steigbild, Strukturen der möglichst ganzen Probe zu erzeugen. Die Methode wurde von Kolisko in den 1929iger Jahren entwickelt, wobei eine Kombination aus Chromatographieprozess und Metallkomplexreaktionen genutzt wurde. Die Firma WALA entwickelte die Methode für die Kontrolle ihrer Produkte und setze Silbernitrat und Eisensulfat ein. Bisher wurde die Methode qualitativ beschreibend ausgewertet, wobei einzelne Bildelemente und deren Interaktion beschrieben wurden. Deshalb musste für die vorliegende Arbeit Auswertungsmethoden entwickelt werden, mit denen auch eine statistische Bearbeitung der Ergebnisse möglich ist (nominale Unterscheidung von proben anhand der Bilder). Die Methode wurde bisher in einer Reihe von Studien eingesetzt (u.a. die Unterscheidung von Produktionsweisen). Obwohl die Bilder nur qualitativ ausgewertet wurden, konnten geschulte Prüfpersonen Proben aus verschiedenen Anbausystemen anhand der Bilder trennen. Die Ergebnisse wurden aber nicht so dokumentiert, dass sie den Erfordernissen internationaler Standardnormen für Laboratorien genügten. Deshalb mussten für diese Arbeit zunächst die Prozeduren dokumentiert und eine systematische Untersuchung zu den Einflussgrößen durchgeführt werden. Dazu wurde die visuelle Bildauswertung entwickelt und standardisiert. Die visuelle Bildauswertung basiert auf morphologischen Kriterien der Bilder von den untersuchten Weizen- und Möhrenproben. Ein Panel aus geschulten Personen entwickelte dann die Kriterien und legte sie anhand von Referenzbildern fest. Die Bilder der vorliegenden Arbeit wurden mit der einfach beschreibenden Prüfung ausgewertet, wie sie aus der sensorischen Prüfung von Lebensmitteln übernommen werden konnte. Mit geschulten und ungeschulten Prüfpersonen wurden Weizenproben und verschiedene Möhrensäfte mit der sog. Dreiecksprüfung ausgewertet (von ISO 4120). Alle Laborprozeduren wurden dokumentiert. Mit der Anwendung dieser Prozeduren wurden Vergleichsversuche mit Laboren in Dänemark und Holland (BRAD, LBI) durchgeführt. Die Ergebnisse waren sowohl für Weizen- als auch für Möhrenproben vergleichbar, wobei alle drei Labore zwischen jeweils zwei Proben unterscheiden konnten. Die systematische Untersuchung zu den Einflussgrößen zeigte, dass das Unterscheidungsvermögen der Methode vor allem von den klimatischen Bedingungen während der Steigphasen beeinflusst wird. Auch die Präkonditionierung der Papiere hat einen großen Einfluss, während die Wasserqualität (ultra-filtriert, de-ionisiert, destilliert) eine untergeordnete Bedeutung hat. Für Weizen- und Möhrenproben wurde sowohl die Wiederholbarkeit als auch die Reproduzierbarkeit getestet. Die Unterschiede in den Bildern der verschiedenen Proben waren dabei immer größer als die Variation durch Proben- und Bildwiederholung und das Labor. Die so charakterisierte Methode wurde auf kodierte Proben von definierten Feldversuchen und auf Marktproben (Paarvergleich von Anbausystemen ökologisch und konventionell) angewandt, wobei als Ergebnis mehr als 90% der Proben mit der einfach beschreibenden Prüfung anhand der Bilder unterschieden werden konnten. Die Auswertung mit der Dreiecksprüfung zeigte, dass sowohl Sorten und Verarbeitungsschritte (Saft) als auch Anbauweisen signifikant getrennt wurden. Darüber hinaus wurde die Methode auch erfolgreich auf Apfelproben angewandt. Weitere Untersuchungen müssen zeigen, ob sich das Potential der Methode, verschiedene Fragen wie die Authentizitätsprüfung von Lebensmitteln verifizieren lassen. Summary (english) Since the 1980s the picture developing methods of food quality determination in organic food have become more important. Picture developing methods include such methods as biocrystallisation, capillary dynamolysis and round-filter chromatographic methods. The results of these methods are patterns, which are evaluated using defined criteria. Biocystallisation shows more or less organised crystal patterns on a round glass plate whereas the patterns from the capillary dynamolysis method are 2-dimensional structures on paper. In the past the patterns resulting from these methods have needed to be evaluated by well trained personnel using product specific criteria. In contrast to thin-layer chromatography, where the compound is separated from the matrix and other substances, the aim of the capillary dynamolysis is to produce structured patterns of the whole juice or extract. The capillary dynamolysis method was originally discovered by Kolisko in the 1920s, where a combination of the capillary analysis and metal-salt-solutions were used. The WALA company applied the method for quality control of their pharmaceuticals. For this they used AgNO3 and introduced FeSO4 to increase the number of form elements as well as to fix the paper. Before the work reported in this paper was undertaken pictures resulting from the above methods had been evaluated qualitatively, describing the different form elements and their interactions. Therefore an evaluation tool had to be developed which allowed a statistical test of the results (nominal differentiation). The capillary dynamolysis method was applied to several different studies for comparison (e.g. farming systems). Although the pictures were only evaluated qualitatively, trained personnel could successfully discriminate samples from different farming systems. However, these results were never documented in a way which met the international standard norms for laboratory methods. Therefore the procedures for these method had to be documented and a systemic investigation of the different influencing factors had to be undertaken. Moreover a visual evaluation method had to be developed and standardised. These tasks are the topic of this Thesis. The visual evaluation of the patterns concerns different morphological criteria for the wheat and carrot samples. A panel of trained assessors was formed and in a consensus meeting all criteria were discussed, agreed and defined. The pictures are evaluated by simple descriptive tests, which were already developed and applied in sensory testing. Moreover the patterns of the wheat samples (varieties) and the carrot juices were evaluated by untrained assessors in a triangular test which was adopted from sensory analysis (ISO 4120). All laboratory procedures were documented according to international standard norms. Using these procedures comparative studies were undertaken between three partner laboratories (Biodynamic Research Association, University of Kassel, Louis Bolk Institute) that resulted in comparable patterns for wheat (variability higher) and carrot samples. Moreover the discrimination ability of the 2 different product types was the same in all laboratories. A systemic investigation of the different factors influencing the patterns was carried out. The discrimination ability between the different wheat and carrot samples is mainly influenced by the climate conditions during the rising phases. Also the preconditioning of the paper material alters the differentiation between the samples. However water from different cleaning systems (ultra-filtration, de-ionization, distillation) does not seem to have an influence. For both samples types (wheat, carrots) the method parameters, repeatability, intermediate precision and reproducibility was tested. The differences between the patterns of the pictures from all samples were greater than the variation from the parallel repetition and the laboratory influence. When the method was applied on coded samples, from defined field trials and from the market samples (pair-wise comparison of organic and conventional farms) more than 90% could be differentiated when the simple descriptive test was applied. When the triangular test was used for the differentiation of the wheat and carrot samples (varieties), carrot juices and carrot samples from different farming systems highly significant results could be achieved. The method was also successfully applied to apple samples from different farming systems. Further investigations need to be carried out to verify the ability of the method to answervarious questions such as the authenticity test of organic crop products for classification. Streszczenie (polski) Od momentu pojawienia sie na rynku produktow rolnictwa ekologicznego, naukowcy staraja sie ustalic roznice jakosciowe pomiedzy produktami ekologicznymi a konwencjonalnymi. Badania jakosci zywnosci odbywaja sie przy zastosowaniu metod analitycznych. W ostatnich latach na znaczeniu zyskuja rowniez tzw. metody holistyczne, do ktorych naleza m.in. metody obrazowania: biokrystalizacja oraz chromatografia obrazowa. Wyniki tych metod to wzory oceniane przy uzyciu zdefiniowanych kryteriow. W przypadku krystalizacji wzory te to skrystalizowane na szklanej plytce substancje. Podczas chromatografii obrazowej na papierze chromatograficznym powstaja wzory dwuwymiarowe. W porownaniu z chromatografia cienkowarstwowa, ktorej celem jest wyizolowanie poszczegolnych substancji z badanej proby, celem chromatografii obrazowej jest uzyskanie odwzorowania czyli wzoru charakterystecznego dla badanej proby. Metoda chromatografii obrazowej odkryta zostala przez Kolisko na poczatku minionego wieku. Kolisko zmodyfikowala metode chromatografii bibulowej m.in. poprzez zastosowanie roztworow soli metali. Firma WALA zastosowala te metode do kontroli jakosci surowcow farmaceutycznych. Jako odczynnikow chemicznych uzyto AgNO3 i FeSO4. Wprowadzenie FeSO4 umozliwilo uwidocznienie wiekszej liczby elementow we wzorach i ich utrwalenie.
